S-300 is a multi layer air defense system that uses 4 missiles, each one with a particular range (each one covers a layer), range up to 300 km and also can intercept short range ballistic missiles. The S-300 is a battery.
The S-22 is usually knows as the Pantzyr S-1 is a short range missile system, its a anti missile defense system to protect high value targets. The Pantzyr S-1 protects the S-300 from missile attacks that evaded the S-300 defenses (like in saturation attacks, etc). They work together.
(Also for other systems: parachute dynamics are worked out very carefully. There is ground information as to wind and other weather conditions.)
For want of anything better at the moment –
For ground penetration I would guess the rocket/bomb needs to fire straight down.
Without the parachute, the bomb would need to be dropped from a greater height and from a further distance due to the ballistic curve.
The distance it takes the parachute to stop the forward momentum at any given release speed would be known. Purely hypothetical figures but say at Mach 1 the bomb is stopped in exactly 500 meters – the bomb would be released 500m before the target.
Retarding the bomb with the parachute would allow the bombing from lower altitudes at greater speeds.
I would guess that these would be used against a complex where the entrances are known but the underground workings are not mapped, so the suspect area would need to be covered in a grid pattern.
Most likely one bombing run would involve dropping a line of bombs (200m ?) on the area known to have underground workings. Next bombing run would lay the next line and so forth.

I don’t think there is any no fly zone as such. I have seen nothing solid on S-300s or S-400s being deployed on land. Only Pantsir which is a short range system. The ships may be able to cover most of Syrian airspace and the US has to be on best behavior when it bombs excavators in ISIS country.
No doubt there would be a no fly zone over Syrian government held territory.
